Is it possible to start a culture from the Rotifer Diet that came with my live rotifers? If it is possible, is there any reason that I should not go this route but instead order the disks and follow the directions on the main website?

The rotifer diet has very little viable plankton in it (if any). You will be much happier and have an easier time of it if you just buy the plankton disks as noted on the main site.
